The High Throughput Screening and Assay Development Facility

Welcome to BIOSS High Throughput Screening Laboratory. Our HTS facility is equipped with a Liquid Handling System, microplate dispenser, microplate washer, microplate reader and RT-PCR cycler. We provide service for liquid handling, screening using the detection modes absorption, fluorescence, luminescence, FRET, TR-FRET and fluorescence polarization and automated Real Time PCR measurements. The liquid handling system is able to pipet the volume of 1 – 1000 µl with high accuracy.

Liquid Handling

Our Tecan system can prepare automatically up to 10,000 samples per day. The system has a LiHa arm that can be connected to fixed tips or disposable tips, depending on the need. An addition of Robotic Arm brings the plates to the shaker, incubator or to the final storage carousel.

Assay Development and Optimization

We offer a service to develop the assay and optimize it to get better assay dynamics and save reagent expenses.

Assay Screening

You can provide us with your assay protocol, and we can screen the assays together. Or, we can screen, process the data and report the Hits for you.

Follow-up

After the screening, we can provide the follow-up as Hit validation, for example Binding constant, IC50, Ki, etc.

LIST OF METHODS

1. Automatisation for library splitting

Pipetting  and diluting up to 10,000 samples per day. The uses of disposable tips allow the pipetting of cell cultures and solutions without cross-contamination.

2. Automatisation for assay screening

The assay screening is performed automatically using liquid handling system (Tecan) and dispenser (Biotek). A washing step can be done with washer (405TS, Biotek). Microplate reader (Synergy H4, Biotek) can monitor the detection via absorption, fluorescence, FRET, TR-FRET and fluorescence polarization.

3. Automatisation for RT-PCR

Pipetting and plate loading are accommodated by the liquid handling system (Tecan) and Robotic arm (Biorob).

4. Assay development

  • Develop and optimize enzyme coupled assay, monitored by absorption, fluorescence, luminescence.
  • Enzyme-linked immunosorbent assay (ELISA)
  • Homogeneous ligand binding assay (fluorescence polarization)
  • Fluorescence Resonance Energy Transfer (FRET)
  • Multiplexing

Tecan Freedom Evo 200 Liquid Handling System

Robotic system with 8 chanel LiHa, heated incubator to 37 °C and shaker, connected with FL601 Recirculating Cooler: -20 - +40 °C

Pipetting: 1-1000 µl

Carousel with stacking system for 220 plates.

 

Biotek Synergy H4 Hybrid reader

96 well microplate/2 min

Detection:

  • Absorption: 96, 384, 1536 well plates
  • Fluorescence: 96 and 384 well plates
  • Luminescence: 96 and 384 well plates
  • FRET

  • Fluorescence Polarization (Excitation: 360/40, 485/20, 540/25 and Emission: 460/40, 528/40, 620/40)
 

Biotek Multiflo Microplate Dispenser

The dispenser can be used for 96, 384 and 1536 microplates, using two different reagents. The machine can dispense 384 well plate in 6 seconds with minimum volume of 1 µl.

Biotek Microplate Washer 405TMTS

The washer accommodates 96 and 384 well plate and can be run alone or automatically through Tecan software. It is ideal for ELISA test and cell based assays.

Lightcycler®480 Real-time PCR

Using Lightcycler RT-PCR, HTS of PCR reactions can be performed to measure the amplification using end-point analysis and melting curve. We accommodate automatisation capability in connection with Tecan liquid handling for pipetting and plate loading.

FL601 Recirculating Cooler

The chiller can be connected to liquid handling system with working temperature between -20 and +40 °C.
